The manuscript presents Aurélien Sérandour's scientific career structured around the causal analysis of the molecular mechanisms involved in pathological conditions. The work is part of an integrative approach aimed at linking chromatin organization, transcriptional regulation, and cellular phenotypes.
The first area of research is devoted to the study of the epigenome, and in particular the hydroxymethylome, as a functional marker of transcriptional regulation. The development and application of high-resolution mapping methods have made it possible to characterize the role of 5-hydroxymethylcytosine in the identification of active regulatory regions and in the structuring of transcriptional programs, particularly in multiple myeloma.
A second area of research focuses on the mechanistic analysis of interactions between chromatin and transcription factors, using approaches combining ChIP-seq, ChIP-exo, and proteomics, which make it possible to link chromatin organization and transcriptional control.
Finally, this manuscript develops functional genetics approaches based on CRISPR screening applied to infectious and tumor contexts, in order to identify genes and pathways involved in cellular behavior. All of this work demonstrates a coherent scientific trajectory and the acquisition of complete autonomy in conducting research projects.
